Cell Quality Assurance:

All the isolated endothelial cells (ECs) and smooth muscle cells (SMCs) are tested by their marker protein expression (Table 1) via both immunocyto chemistry (Figure A & B) and FACS analysis (Figure C) to assure the quality. Among tested cells > 90% marker positive cell colonies are distributed to the requested researchers.

Table 1: Quality Control Markers for Smooth Muscle Cells and Endothelial Cells

Endothelial cell marker immunostaining. Left: CD31 immunostaining (green) of Type II (intermediate size PA) endothelial cells isolated from IPAH patients. Right: vWF immnostaining (green) of Type III (pre-capillary PA) endothelial cells isolated from IPAH patients. DAPI (blue) is used for nuclear counter staining

Smooth muscle marker immunostaining. Left: a-SMA, Sm22a (middle) and SMMHC (right) immunostaining (green) of Type II (intermediate size PA) SMC isolated from IPAH patients. DAPI (blue) is used for nuclear counter staining

Endothelial cell marker FACS analysis. Left: no staining counting for background threshold determination. In this case < 104 is the threshold. Right: CD31-FITC FACs analysis indicating that over 95% of the cells are CD31 positive
